Ictotest tablet test (Ames) -The ictotest tablet test is used to
confirm positive reagent strip test since it is more sensitive than all of
them. The test is a quick, simple and standardised colour test.
Principle- Bilirubin in urine reacts with a
solid diazonium salt in the presence of salicylsulphonic acid to form a blue or
purple colour. The tablet contains p-nitrobenzene diazonium p-toluene
sulphonate, salicylsulphonic acid and sodium hydrogen carbonate.
Procedure-- 1. Place 5 drops of urine on a
special test mat supplied with the kit.
2. Place a tablet in the middle of
moist area on the mat.
3. Add 2 drops of water on the
tablet.
4. Observe the colour of the mat
within 30 seconds.
Results- No colour change around the tablet on
the mat, or there is slight red or pink within 30 seconds is a negative test.
Colour change to bluish purple around the tablet indicates a positive test. The
concentration of bilirubin in the urine is pro portional to the intensity of
the colour and rate at which colour is developed.
